The gBlock provided by IDT was perfect and the protocol provided on the main page worked so we were able to make an sRNA fusion to target expression of the fliC gene from S. marcescens (BBa_K1963011). The fliC target, encoding flagellin, was chosen as a Proof-of-Concept, since successful sRNA inhibition should affect swimming, which can be tested in the laboratory, but the flagellum is also important for pathogenesis in many bacteria. In the S. marcescens DB10 model system, which is an insect pathogen that is not a biohazard for laboratory use, the FliC protein is doubly useful as it is readily seen in culture supernatants separated by SDS-PAGE and stained with Coomassie Blue.
